Purpose/Objective(s) Recurrence and metastasis of nasopharyngeal carcinoma (NP) is closely related to immune escape. It known that LMP1 promotes the escape cells (NPCs) by controlling expression PD-L1. However, underlying molecular mechanisms remain be fully characterized. Materials/Methods At first, we screened transcription factor which can induce, find regulates PD-L1, further proved elevated IRF-1 in cells. We confirmed presence LMP1/Lgals1 complex, demonstrated regulate through NF-κB signaling pathway. also involvement Lgals1 T-cell killing assay, ELISA flow cytometry. Results In this study, interacts with Lgals1, a beta (β)-galactoside-binding protein, IRF1 via NF-kB pathway, promoting PD-L1 NPCs. are both found tightly linked recurrence NPC based on clinical data. Targeting using OTX008, specific inhibitor, able restore immunogenic environment Conclusion Our findings reveal regulatory axis for programmed death ligand 1 (PD-L1) cells, targeting one component axis, effective boosting immunogenicity NPCs, providing therapeutic avenue treating clinic.
